The purpose of present study was to compare and evaluate the effect of layer-to-layer technique of autogenous bone and deproteinized bovine bone and a mixture graft of autogenous bone and DBB on exophytic bone formation in rabbit calvaria. TR e-PTFE membrane was contoured in rectangular parallelepiped shape (8 ㎜ length, 5 ㎜ width and 4 ㎜ inner height). Thirty-two adult male New Zealand white rabbits (mean weight 3 ㎏) were used in this study and 8 defects were allotted to each test group randomly. Decortication was performed on calvaria to promote blood supply in wound area and membrane was fixed with 4 fixation pins at each surgical site. The test groups were divided into 3 as follows : TR + Autograft(I) + DBB(O) (TestⅠ), TR + DBB(I) + Autograft(O) (TestⅡ), TR + Mixed (Autograft+DBB) (Test Ⅲ), TR only was served as control. The experimental animals were sacrificed at 4 and 8 weeks after surgery as scheduled. Non-decalcified resin embedded preparation was processed for histologic and histomorphometric analysis. And statistical analysis was performed for assessment of new bone formation, bone fill. The results of this study were as follows: 1.New bone formation was increased in control group at 8 weeks than 4 weeks and the difference was significant (P<0.05). 2. Control group showed greater new bone formation than test groups at 4 and 8 weeks. 3. Test group II showed greater new bone formation than other test groups at 8 weeks although the difference was not significant. 4. In the top area of membrane space (section I), test group II showed more new bone formation than other groups at 4 weeks and the difference between test group II and control group was statistically significant(p<0.05). Test group II showed more new bone formation than other groups at 8 weeks and the differences between test group II and control group, between test group II and test group III were statistically significant(p<0.05). 5. In the top area of membrane space (section I), test group II showed more bone fill than other groups at 4 weeks and the differences between test group II and control group, between test group II and test group I were statistically significant(p<0.05). Test group I showed more bone fill than other groups at 8 weeks and the difference between test group I and control group was statistically significant(p<0.05). Within the limit of our experiment, it can be concluded that more favorable new exophytic bone formation could be obtained in use of composite graft of autogenous bone and deproteinized bovine bone with layer-to-layer technique (Autogenous bone is located in outer layer).
