Bissuk (virgate wormwood, 豬毛蒿), also known as Artemisia scoparia Waldst. et Kitaib (AS), is a plant that used in pharmaceutical treatment and cosmetic. However, the effect of AS on atopic dermatitis (AD) has not been reported. In this study, the author examined inhibitory effect of AS on AD-like murine model. 2,4-dinitrofluorobenzene (DNFB) was applied in dorsal skin of BalB/c mice to induce AD-like symptoms and AS or AS butanol fraction (Bu-OH) was topically applied for 4 weeks. Results showed that AS and Bu-OH suppressed scratching behavior and clinical symptoms in AD-like skin lesion. Infiltrated inflammatory cells in skin lesions were significantly reduced by AS or Bu-OH application. IgE level in serum of AD-like mice was significantly suppressed by topical application of AS. Bu-OH significantly suppressed levels of histamine, IgE, thymic stromal lymphopoietin (TSLP), interleukin (IL)-4, and IL-6 in serum of AD-like mice. AS and Bu-OH significantly reduced mRNA levels of inflammatory cytokines in skin lesions of AD-like mice. Additionally, caspase-1 activities in AD-like skin lesions were also significantly suppressed by AS and Bu-OH. In conclusion, these results propose therapeutic potential of AS and Bu-OH by showing the scientific evidences on anti-AD effect.

비쑥 (豬毛蒿, Artemisia scoparia Waldst. et Kitaib)은 국화과 쑥속에 속하는 식물로 염증, 발열 및통증 완화 등에 효과를 나타내는 것으로 알려져 있다. 저자는 2,4-dinitrofluorobenzene (DNFB)를 이용해 아토피 피부염이 유도된 마우스모델에 비쑥과 비쑥의 부탄올 분획 (Bu-OH)을 4주간 도포한 후 마우스의 혈청과 등조직에서 ELISA, RT-PCR, caspase-1 assay 등을 시행하여 그 효과를 규명하였다. 그 결과, 비쑥과 Bu-OH는 가려움증을 포함한 임상증상을 완화시켰고, 등조직의 표피두께와 침윤된 염증세포수를 감소시켰다. 비쑥은 마우스 혈청의 IgE를 유의하게 감소시켰고, Bu-OH는 히스타민, IgE, 흉선 기질상 림포포이에틴 (Thymic stromal lymphopoietin, TSLP), IL-4 및 IL-6를 유의하게 감소시켰다. 또한 비쑥은 등조직에서의 종양괴사인자 (tumor necrosis factor (TNF)-α)의 mRNA 발현과 caspase-1의 활성을 감소시켰고, Bu-OH는 TSLP, IL-4, IL-6, TNF-α의 mRNA 발현과 capase-1 활성을 감소시켰다. 이러한 결과를 통해 본 연구는 비쑥과 Bu-OH가 TSLP 생성 조절을 통해 아토피 피부염을 개선하는 효과를 가짐을 시사한다.
